The RNeasy Microarray Tissue Mini Kit provides the optimal solution for purifying RNA for microarray analysis. QIAzol Lysis Reagent ensures efficient lysis of even difficult-to-lyse tissues, while trusted RNeasy spin columns allow fast and convenient purification of intact RNA without phenol contamination. The result is highly pure RNA that delivers reliable results in microarray analysis and avoids the need for unnecessary repetition of experiments. For cultured cells, we recommend using the RNeasy Microarray Tissue Mini Kit or RNeasy Plus Mini Kit for RNA purification. The procedure can be fully automated on the QIAcube.

RNeasy Microarray Tissue procedure.

For the RNeasy Microarray Tissue procedure, there is no need to precipitate RNA prior to RNA cleanup. Purification of RNA from precious samples can be conducted without the risk of failure due to loss of RNA pellets during the purification procedure or due to incomplete removal of contaminants such as phenol.

Intact RNA.
Total RNA was purified from [A] rat kidney and [B] rat brain using the RNeasy Microarray Tissue procedure and then analyzed on the Agilent 2100 Bioanalyzer. The presence of 2 major peaks, representing 28S rRNA and 18S rRNA, indicates that the RNA is intact.

Reliable microarray analysis.
RNA was purified from 13 gliomas (4 with complete 1p19q codeletion, and 9 with EGFR amplification) using QIAzol/RNeasy technology, and microarray analysis was performed using the Affymetrix GeneChip Human Genome U133 Plus 2.0 Expression Array. Unsupervised hierarchical clustering analysis was carried out using the 1366 probe sets whose expression varied the most across all samples. Lowest and highest intensity values are represented by blue and red, respectively. Analysis revealed a clear difference in gene expression between gliomas with 1p19q codeletion and gliomas with EGFR amplification. (Data excerpted from Ducray, F. et al. The RNeasy Microarray Tissue procedure halves the time to purify RNA for microarray analysis.
Performance

RNA purified using the RNeasy Microarray Tissue Mini Kit is highly pure and intact (see figure "Intact RNA"), ensuring reliable results in microarray analysis (see figure "Reliable microarray analysis"). The RNeasy Microarray Tissue procedure is suitable for all tissue types, delivering high yields of RNA from a range of samples, including fibrous and fatty tissues (see table).

Principle

The RNeasy Microarray Tissue Mini Kit, with QIAzol Lysis Reagent and RNeasy spin columns, provides the optimal solution for purifying RNA for microarray analysis. Obtaining sufficient yields of RNA, especially from fibrous and fatty tissues, is a challenge. QIAzol Lysis Reagent efficiently lyses all tissue types, delivering optimium starting material for isolation of RNA for microarray analysis. (For fibrous tissue, proteinase K needs to be purchased separately.) Use of RNA containing trace contaminants affects the reverse-transcription step in microarray experiments. RNeasy spin columns efficiently remove phenol and other contaminants, providing highly pure RNA suitable for sensitive applications like microarray analysis.

Procedure
For the RNeasy Microarray Tissue procedure, tissues are rigorously lysed in phenol-guandine–based QIAzol Lysis Reagent, and RNA is purified from the aqueous phase using an RNeasy spin column (see figure "RNeasy Microarray Tissue procedure"). There is no need to precipitate RNA prior to RNA cleanup, which prevents potential loss of RNA, avoids variable RNA yields, and saves time (see figure "Significant time savings"). For disruption and homogenization of tissues, we recommend TissueRuptor and TissueLyser instruments. Subsequent RNA purification can be automated on the QIAcube.
Applications

The RNeasy Microarray Tissue Mini Kit is suitable for isolating RNA for microarray experiments. Purification of RNA from precious samples can be conducted without the risk of failure due to loss of RNA pellets or incomplete removal of contaminants such as phenol. The purified RNA is also ready to use for other applications, such as real-time RT-PCR.
