The BioRobot Universal System integrates all the instrumentation, software, purification technologies, and enzyme technologies that are required for high-throughput applications. Application Packs are available for gene expression, genotyping, sequencing, forensics, veterinary, and pharma applications. Fully automated, optimized protocols enable purification of highly pure RNA, genomic DNA, and plasmid DNA plus PCR cleanup. Automated RT-PCR, PCR, sequencing reaction, and forensic assay setup provides high levels of reliability and standardization and helps to eliminate variation caused by handling errors, particularly when processing a large number of samples.

Highly reproducible purification of RNA from blood.

Blood was drawn from 15 donors and collected in PAXgene Blood RNA Tubes. Six samples were taken from each donor. Total RNA was purified using the PAXgene Blood RNA MDx Kit on the BioRobot Universal System. The mean RNA yield for each sample was determined spectrophotometrically. The purification procedure showed high intra-donor repeatability.

Reproducible real-time PCR results.
Genomic DNA was purified from 42 buccal swabs using the QIAamp 96 DNA Swab BioRobot Kit on the BioRobot Universal System. Purified DNA was eluted in 100 µl RNase-free water, and 5 µl was used for PCR. Quantitative real-time PCR (25 µl reaction volume) was performed on the ABI PRISM 7700 Sequence Detection System using the QuantiTect Probe PCR Kit with primers and probe for the β-actin gene. The efficiency and reliability of the purification procedure is demonstrated by the reproducible CT values.

Highly reproducible purification of plasmid DNA.
Molecular Grade plasmid DNA was purified from 96 E. coli cultures using the QIAprep 96 Turbo BioRobot Kit on the BioRobot Universal System. The average A260/A280 absorbance reading was 1.87, demonstrating the high purity of the DNA.

Comparable recovery to the manual procedure.

A 500 bp PCR product (3 µg) was purified using the MinElute 96 UF PCR Purification Kit on the BioRobot Universal System or using the manual procedure. DNA was eluted in 30 µl using the automated procedure and in 50 µl using the manual procedure. Recovery using the automated procedure on the BioRobot Universal System was comparable to the manual procedure.

High-quality RNA from adipose tissue.
Total RNA was purified from adipose tissue using [A] the RNeasy 96 Universal Tissue 8000 Kit on the BioRobot Universal System or [B] a manual procedure with the RNeasy 96 Universal Kit. Purified RNA was analyzed on the Agilent 2100 bioanalyzer. The RNA Integrity Number (RIN), indicated on each scan, shows the high quality of the purified RNA.

Linear CT values over a wide dynamic range.
RNA was purified from 10 to 5 x 105 Jurkat cells using the RNeasy 96 BioRobot 8000 Kit on the BioRobot Universal System. Purified RNA was eluted in 100 µl RNase-free water, and 5 µl of a 1:10 dilution of the eluate was used for RT-PCR. Quantitative real-time one-step RT-PCR (25 µl reaction volume) was performed on the ABI PRISM 7700 Sequence Detection System using the QuantiTect Probe RT-PCR Kit with primers and probe for the β-actin transcript. The plot shows linear CT values over a wide range of starting template amounts.

Purification of highly pure RNA from blood.
Blood was drawn from 13 donors and collected in PAXgene Blood RNA Tubes. Six samples were taken from each donor. Total RNA was purified using the PAXgene Blood RNA MDx Kit on the BioRobot Universal System. Absorbance measurements were taken at 260 nm and 280 nm for each RNA sample, and the ratio of the absorbance (A260/A280) was calculated. The values obtained showed that the RNA was highly pure.
Performance
Gene expression applications

The BioRobot Universal System together with the Application Pack, Gene Expression enables walkaway RNA purification from blood (collected in PAXgene Blood RNA Tubes), tissues, and cells plus RT‑PCR setup in 96-well format. The automated purification procedure provides highly pure RNA from blood (see figure “Purification of highly pure RNA from blood”) with high intra-donor repeatability (see figure “Highly reproducible purification of RNA from blood”).

The BioRobot Universal System together with the RNeasy 96 Universal Tissue 8000 Kit enables purification of high-quality RNA from tissues — including difficult‑to‑lyse fatty (see figure “High-quality RNA from adipose tissue”) and fibrous tissues.

RNA can be purified from cells using the RNeasy 96 BioRobot 8000 Kit. The reliability and efficiency of the purification procedure is demonstrated by linear CT values over a wide range of starting template amounts, from as few as 10 cells up to 5 x 105 cells (see figure “Linear CT values over a wide dynamic range”).

Genotyping applications

The BioRobot Universal System together with the Application Pack, Genotyping enables walkaway purification of genomic DNA from buccal swabs or human whole blood. High-quality genomic DNA can be purified from up to 192 buccal swab samples or up to 96 human whole blood samples using a fully automated protocol. The automated purification procedure completely removes all contaminants and enzyme inhibitors and purified DNA is ready for immediate use in high-throughput genotyping applications. Highly reproducible CT values are achieved (see figure “Reproducible real-time PCR results”), demonstrating the efficiency and reliability of the purification procedure.

Automated PCR setup provides high levels of reliability and standardization and helps to eliminate variation caused by handling errors, particularly when processing a large number of samples. With the BioRobot Universal System, highly reproducible CT values are achieved — with no detectable cross-contamination (see table).

Amplification reactions were set up using an automated procedure on the BioRobot Universal System. Columns of a 96-well plate contained either RNAse-free water (1, 3, 5, 7, 9, and 11) or 4 ng template DNA (2, 4, 6, 8, 10, 12) in an alternating sequence. Real-time PCR was carried out using the QuantiTect Probe PCR Kit and primers and probe specific for the β-actin gene. CT values are indicated. X: No signal was detected after 35 cycles.

Sequencing applications

The BioRobot Universal System together with the Application Pack, Sequencing enables walkaway purification of plasmid DNA, DNA cleanup, and sequencing reaction setup. Using the QIAprep 96 Turbo BioRobot Kit, Molecular Biology Grade plasmid DNA can be purified from up to 4 x 96 cultures of E. coli (1–5 ml) per run. DNA yields are highly reproducible (see figure “Highly reproducible purification of plasmid DNA”).

PCR products (>100 bp) can be purified from up to 4 x 96 samples per run using a fully automated protocol with the MinElute 96 UF PCR Purification Kit. Recovery rates of PCR products are comparable to the manual procedure (see figure “Comparable recovery to the manual procedure”), demonstrating the reliability of the automated procedure.

Investigator applications

The BioRobot Universal System together with the Application Pack, Investigator enables purification of total DNA from a range of forensic casework samples including dried blood, surface swabs (e.g., blood, semen, saliva, and epithelial cells), stains on fabric (e.g., blood- or saliva-stained fabrics), cigarette butts, tobacco, chewing gum, and envelopes.

Using proven QIAamp 96 technology, purification is efficient and purified DNA performs well in sensitive downstream analyses, such as STR analysis, with high hit rates. The robust purification procedure, using the QIAamp 96 DNA Swab BioRobot Kit, provides high process safety and helps to prevent cross-contamination.

Principle

The BioRobot Universal System enables fully automated purification of high-quality nucleic acids from a variety of sample types. In addition, the instrument can perform downstream reaction setup in 96-well format. By automating nucleic acid purification and reaction setup, the BioRobot Universal System saves hands-on time and ensures an efficient, streamlined workflow.

Purification and downstream reaction setup are provided by each Application Pack. Four Application Packs are available:

Procedure

Automated purification on the BioRobot Universal System follows a simple bind, wash, elute procedure. The user selects the purification protocol and software messages guide the user through the remaining steps required to set up the instrument. The number of samples per run depends on the sample type and the analyte purified (see table).

QIAsoft 5, the controlling software for the BioRobot Universal System, enables easy system operation and reproducible sample processing. A detailed load check ensures that samples, reagents, and labware are correctly loaded, helping to eliminate human error during worktable setup. QIAsoft 5 enables easy administration of users, and report and result files are protected using Checksum to detect inappropriate modification.

QIAsoft 5 also enables users to comply with the technical requirements of 21 CFR Part 11. In addition, files used or generated by QIAsoft 5 enable easy data management and compatibility with laboratory information management systems (LIMS). The spreadsheet functionality within QIAsoft 5 enables easy setup of pipetting schemes for customized applications.
