Identification of miRNA regulators of INPPL1 expression.

PCR reactions using primers for INPPL1 (SHIP2) and a normalization gene were performed using the SureFIND Cancer miRNA Transcriptome PCR Array. The SureFIND Cancer miRNA Transcriptome PCR Array contains cDNA samples derived from HeLa cells treated with 90 different cancer-associated miRNA mimics. The software identified miRNA-205, –193b, –138, –373, –29b, and –135b as negative regulators of INPPL1 expression.

Performance

Principle

SureFIND Transcriptome PCR Arrays are innovative tools for identifying which miRNA, signaling pathway, or transcription factor regulates the expression of any gene.

In each well of a SureFIND Transcriptome PCR Array is a unique "transcriptome" or cDNA sample synthesized from cells treated with a unique miRNA mimic, chemical inhibitor, or siRNA. Using real-time PCR, researchers can identify the mechanisms of gene expression regulation. A change in the expression of the tested gene in a specific sample shows that the corresponding miRNA, signaling pathway activity, or transcription factor is a regulator of that gene.

Procedure

Using SureFIND miRNA Transcriptome PCR Arrays is as simple as setting up a real-time PCR experiment. Simply mix primers with the appropriate PCR master mix, aliquot equal volumes into each well of the plate, and run the plate in a real-time PCR instrument (see figure "SureFIND Transcriptome PCR protocol").

After running the PCR, the Excel-based data analysis template for SureFIND Transcriptome PCR Arrays performs the ∆∆CT-based fold-change. The results are delivered in a tabular format and assists in hit selection.
