Performance
flocktype Mycoplasma Mg Ab is specific for the detection of antibodies to Mycoplasma gallisepticum. Tests were run using the flocktype Mg Ab Kit in the Deventer International Proficiency Testing Scheme (PTS) ring trial. Sample sets were from either vaccinated chickens (samples 3 and 6) or from infected chickens (samples 1, 2, 4, 7, and 8). Three positive serum samples were correctly scored as being positive for Mg antibodies (samples 2, 3, and 5). Samples 1 and 7 were Ms-positive with high levels of antibodies to Mycoplasma synoviae. They show low cross reactivity with flocktype Mycoplasma Mg Ab giving doubtful (sample 1) or a low positive signal (sample 7). Samples 4 and 8 (Mycoplasma meleagridis; Mm) were correctly scored as negative. Sample 6 is taken from SPF layers that had been vaccinated with a Nobilis Mg S6/85 vaccine. This vaccine does not induce an antibody response and therefore is correctly scored as negative (see the table Deventer ring trial Mycoplasma Mg 2012 results).

Principle

Mycoplasma gallisepticum is found worldwide and causes chronic respiratory diseases, decreased growth, and reduced egg production in birds. Morbidity and mortality can vary widely and depend on stress conditions and secondary infections.

Mycoplasmas adhere to the respiratory epithelium and to erythrocytes. The cellular immune response is central to infections with M. gallisepticum, although the production of specific antibodies may also be induced by infections and vaccinations. For confirmation of the flocktype Mycoplasma Mg Ab results, we recommend the identification of the pathogen from tracheal swabs using PCR (e.g., with the bactotype Mycoplasma Mg/Ms PCR Kit).

flocktype Mycoplasma Mg Ab is an ELISA (enzyme-linked immunosorbent assay) with ready-to-use colored reagents. The test plate is coated with a recombinant membrane protein that is specific for M. gallisepticum. This protein is highly conserved amongst the M. gallisepticum strains and is strongly immunogenic.

During the sample incubation M. gallisepticum-specific antibodies bind to the immobilized antigen. The anti-IgY-HRP conjugate detects antibodies bound to the antigen. The color reaction is started by adding the substrate solution and stopped after 10 minutes. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of anti-M. gallisepticum antibodies in the sample.

 

Procedure

Serum and plasma samples can be tested after dilution.

Samples are incubated for 30 minutes with the test plate and if antibodies to M. gallisepticum are present, they will bind to the antigen on the plates. Unbound material is removed by rinsing.

The polyclonal anti-IgY-HRP conjugate is added. Unbound conjugate is rinsed out.

The colour reaction is started by adding the TMB Substrate Solution and stopped after 10 minutes. The optical density (OD) is measured using a spectrophotometer. The results are easy to interpret.
