The bactotype Mycoplasma Mg/Ms PCR Kit allows the safe and simultaneous detection of DNA from Mycoplasma gallisepticum and M. synoviae in samples from poultry. The kit contains all of the reagents for multiplex real-time PCR in a single ready-to-use reaction mix. It can be used with individual and pooled samples from tracheal and oropharyngeal swabs and culture medium.

Using the bactotype Mycoplasma Mg/Ms Kit, real-time PCR was performed in triplicate on a titration series of in vitro DNA (2 x 105 copies/μl). There is a linear correlation between the log of the DNA copy number and the amplification results (CT). The correlation coefficient is 0.997.

Using the bactotype Mycoplasma Mg/Ms Kit, real-time PCR was performed in triplicate on a titration series of in vitro DNA (2 x 105 copies/μl). There is a linear correlation between the log of the DNA copy number and the amplification results (CT). The correlation coefficient is 0.997.

Performance

bactotype Mycoplasma Mg/Ms PCR Kit simultaneously detects DNA from Mycoplasma gallisepticum (Mg) and Mycoplasma synoviae (Ms) in samples. A validation study of 450 samples (kindly provided by the Friedrich-Loeffler-Institute and private laboratories) with known Mycoplasma status was tested with the bactotype Mycoplasma Mg/Ms PCR Kit. Of 284 positive samples, 280 samples tested positive, providing a diagnostic sensitivity of 98.6%. All negative samples in this study tested negative, providing a diagnostic specificity of 100%. In addition, a titration series was performed with ten-fold dilutions from 106 to 1 copy/well of in vitro Mg and Ms DNA. The results demonstrate a high correlation (99%) between the number of copies and the amount of amplification product in the range from 106 to 10 copies/well. Ms DNA was detectable down to 10 copies/well and Mg DNA down to 1 copy/well (see the figures Sensitive detection of Mycoplasma Mg and Sensitive detection of Mycoplasma Ms). 

Principle
This is a real-time multiplex PCR kit for the detection of DNA from Mycoplasma gallisepticum and M. synoviae. PCR is based on the amplification of specific regions of the pathogen genome. It is a sensitive and specific tool for pathogen detection.

Real-time PCR involves the amplification of specific DNA by PCR. The amplified product is detected using fluorescent dyes. These are usually linked to oligonucleotide probes that bind specifically to the amplified product. Monitoring the fluorescence intensities during the PCR run (i.e., in real time) allows the detection of the accumulating product without the need to re-open the reaction tubes afterward.

The kit contains all the necessary reagents for the assay, including positive and negative controls. Three specific primer/probe combinations are used: one for M. gallisepticum, one for M. synoviae, and one for the internal control.

Procedure
The kit contains a ready-to-use mix with all of the necessary reagents for the detection of DNA from M. gallisepticum and M. synoviae and the required positive and negative controls.

DNA is extracted from the starting material (tracheal and oropharyngeal swab samples or culture medium) using a suitable sample preparation kit. The extracted DNA is used for the PCR, which is run on a thermal cycler.

The bactotype Mycoplasma Mg/Ms PCR Kit is for detection of DNA from Mycoplasma gallisepticum and M. synoviae in animal samples.
