QIAprep 96 Plus Kits enable plasmid preparation in 96-well format. Samples can be processed using QIAvac 96 and/or a centrifuge or automated on the BioRobot Universal System. Up to 50 μg of high-quality plasmid DNA (endotoxin levels <10 EU/μg DNA) can be obtained from up to 5 ml of culture.
For optimal results it is recommended to use this product together with QIAvac 96.

High yields of plasmid DNA.
E. coli DH5a strains carrying pCMVβ were grown in 5 ml LB medium. Plasmid DNA was isolated manually using the QIAvac 96 and either the QIAprep 96 Plus Miniprep Kit or the QIAprep 96 Turbo Miniprep Kit or automated on the BioRobot Universal System using either the QIAprep 96 Plus BioRobot Kit or the QIAprep 96 Turbo BioRobot Kit. DNA was quantified using absorbance measurement and verified by agarose gel analysis. Higher yields were achieved with QIAprep 96 Plus Miniprep and BioRobot Kits.

Superior performance compared to other suppliers.
E. coli DH5a strains carrying pCMVβ were grown in LB medium. Preps were performed according to the manufacturer's instructions. DNA was quantified using absorbance measurement and verified by agarose gel analysis. The QIAprep 96 Plus Miniprep Kit provided the highest yields compared to the other kits tested.

Superior plasmid DNA quality.
pCMVβ DNA was prepared using the QIAprep 96 Plus Miniprep Kit and kits from other suppliers. For agarose gel analysis, 150 ng (as determined by absorbance measurement) of each sample was used. With the QIAprep 96 Plus Miniprep Kit, a high percentage of supercoiled plasmid DNA (lower band) was observed and absorbance measurements and agarose gel analysis were comparable, indicating superior plasmid DNA quality. M: GelPilot 1 kb Plus Ladder (the most intense fragment corresponds to 100 ng).
Performance
High-purity preps and outstanding yields

QIAprep 96 Plus Kits provide a novel method for preparation of highly pure DNA (see figure "Superior plasmid DNA quality"). Yields of up to 50 μg can be consistently achieved using an optimized procedure. The unique binding buffer provided with the kits ensures very specific binding conditions (see figures "High yields of plasmid DNA" and "Superior performance compared to other suppliers").

Principle
The unique kit chemistry and design of the Plasmid Plus 96 plates allow high binding capacities. After neutralization, a TurboFilter 96 plate is used for lysate clearing. To optimize plasmid DNA binding to the membrane of the Plasmid Plus 96 plate, a nonchaotropic binding buffer is added to the cleared lysate. The unique binding buffer ensures very specific binding conditions, resulting in DNA quality that is comparable to anion-exchange preps (see figure "Superior plasmid DNA quality"). Due to unique kit chemistry, up to 50 μg plasmid DNA per well can be obtained from a 5 ml E. coli culture (see figures "High yields of plasmid DNA" and "Superior performance compared to other suppliers").
Procedure

QIAprep 96 Plus Kits allow plasmid preparation in 96-well format. The QIAprep 96 Plus Miniprep protocol is based on a modified alkaline lysis procedure. Following lysate clearing, a simple bind-wash-elute procedure results in highly concentrated plasmid DNA, ready for direct use in subsequent applications (see flowchart "QIAprep 96 Plus Miniprep procedure").

Convenient and flexible processing

The QIAprep 96 Plus Miniprep Kit can be conveniently processed using the QIAvac 96 and/or a centrifuge, while preps can be automated on the BioRobot Universal System using the QIAprep 96 Plus BioRobot Kit. Optimized protocols ensure easy and straightforward plasmid DNA preparation in 96-well format.
