Easy-to-use, the AllPrep Bacterial DNA/RNA/Protein Kit isolates total nucleic acids and cellular proteins from bacterial cultures using spin column technology. Using a bead-based lysis and spin-column collection method, nucleic acids and proteins are effectively extracted from Gram (+/-) bacterial species. DNA and RNA is ready for qPCR, RT-PCR and next-generation sequencing, while protein is ready for 1D SDS-PAGE and mass spectrometry following in-gel trypsin digestion. Some applications, including 2D SDS-PAGE and solution phase proteolytic digestion for mass spectrometry, may require an additional detergent removal step following protein elution.
Beginning by lysing cultured bacterial cells using 0.1 mm glass bead tubes, nucleic acids and proteins are made soluble and bound with solution. In three steps of binding and flow through, DNA is bound to one silica spin column and the flow-through containing RNA and proteins is processed through a second and third spin column using buffers designed to assist binding each component. Each spin column is then individually washed and immobilized analyte is eluted.
