flocktype Mycoplasma Ms Ab is specific for the detection of antibodies to Mycoplasma synoviae. Tests were run using the flocktype Ms Ab Kit in the Deventer International Proficiency Testing Scheme (PTS) ring trial. Sample sets were from either vaccinated chickens (samples 3 and 6) or from infected chickens (samples 1, 2, 4, 7 and 8). Four positive serum samples were correctly scored as being positive for Ms antibodies (samples 1, 4, 5, and 7). Samples 3, 6, and 8 [Mg- or Mycoplasma meleagridis (Mm)-positive] correctly scored as negative. Sample 2 is an Mg-positive serum that gave a doubtful result with flocktype Mycoplasma Ms Ab (see the table Deventer ring trial Mycoplasma Ms 2012 results).

Mycoplasma synoviae is found worldwide and causes chronic respiratory diseases in birds. Systemic infection leads to infectious synovitis. The severity of infection depends on secondary infections.
Mycoplasmas adhere to the respiratory epithelium and to erythrocytes. The cellular immune response is central to infections with M. synoviae, although the production of specific antibodies may also be induced by infections and vaccinations. For confirmation of the flocktype Mycoplasma Mg Ab results, we recommend the identification of the pathogen from tracheal swabs using PCR (e.g., with the bactotype Mycoplasma Mg/Ms PCR Kit).
flocktype Mycoplasma Ms Ab is an ELISA (enzyme-linked immunosorbent assay) with ready-to-use colored reagents. The test plate is coated with a recombinant membrane protein that is specific for M. synoviae. This protein is highly conserved amongst the M. synoviae strains and is strongly immunogenic.
During the sample incubation M. synoviae-specific antibodies bind to the immobilized antigen. The anti-IgY-HRP conjugate detects antibodies bound to the antigen. The color reaction is started by adding the substrate solution and stopped after 10 minutes. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of anti-M. synoviae antibodies in the sample.
Serum and plasma samples can be tested after dilution.
Samples are incubated for 30 minutes with the test plate and if antibodies to M. synoviae are present, they will bind to the antigen on the plates. Unbound material is removed by rinsing.
The anti-IgY-HRP conjugate is added. It contains an HRP-labeled, anti-IgY-specific polyclonal antibody. Unbound conjugate is rinsed out.
The colour reaction is started by adding the TMB Substrate Solution and stopped after 10 minutes. The optical density (OD) is measured using a spectrophotometer. The results are easy to interpret.
