Performance
flocktype IBV Ab is specific for the detection of antibodies to infectious bronchitis virus. Tests were run using the flocktype IBV Ab Kit in the Deventer International Proficiency Testing Scheme (PTS) ring trial. Sample sets were from either vaccinated chickens (samples 1, 5, 6, and 7) or from infected chickens (sample 3). Four positive serum samples were correctly scored as being positive for IBV antibodies (samples 3, 5, 6, and 7). Serum sample 8 is taken from commercial layers that had been vaccinated in the rearing period with live and inactivated IBV vaccines and therefore correctly tested positive. Due to the early phase of vaccination sample 1 provides a negative result. The two SPF sera (samples 2 and 4) were correctly scored as negative (see the table Deventer ring trial IBV 2007 results).

The kit correctly scored 7 of the 8 samples. Due to the early phase of vaccination, sample 1 tested negative for IBV-specific antibodies. 
Principle

The clinical signs of infectious bronchitis depend on the age and immune status of the chicken, the transmission route, and the virus strain. In chicks between 1 and 4 weeks of age, gasping, coughing, rale, nasal exudate, and respiratory distress can be observed. The mortality rates are normally between 25 and 30%.

flocktype IBV Ab is an ELISA (enzyme-linked immunosorbent assay) with ready-to-use colored reagents. The test plate is coated with a recombinant nucleocapsid protein from the virus. This protein is highly conserved amongst infectious bronchitis virus strains and it is considered to be a major immunogenic peptide.

During the sample incubation, infectious bronchitis virus-specific antibodies bind to the immobilized antigen. The anti-IgY-HRP conjugate detects antibodies bound to the antigen. The color reaction is started by adding the substrate solution and stopped after 10 minutes. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of anti-infectious bronchitis virus antibodies in the sample.

Procedure

Serum and plasma samples can be tested after dilution.

Samples are incubated for 30 min with the test plate. If antibodies to infectious bursal disease virus are present, they will bind to the antigen on the plates. Unbound material is removed by rinsing.

The anti-IgY-HRP conjugate is added. Unbound conjugate is rinsed out.

The colour reaction is started by adding the TMB Substrate Solution and stopped after 10 minutes. The optical density (OD) is measured using a spectrophotometer. The results are easy to interpret.
