Principle

Cignal Lenti Reporter Assays are ready-to-transduce lentiviral particles for assessing cell signaling activities in virtually any mammalian cell type. Cignal Lenti Reporter Assays utilize a unique combination of transcription factor reporter technology coupled with lentiviral delivery.

Positive Control (luc) CMV enhancer/promoter driven firefly luciferase gene with puromycin selection marker. Measure transduction efficiency and serve as positive control for firefly luciferase assay
Negative Control (luc) Minimal promoter driven firefly luciferase gene with puromycin selection marker. Establish the specificity of any treatment effects and determine background firefly luciferase activity
CMV-Renilla Control (luc) CMV enhancer/promoter driven Renilla luciferase gene with puromycin selection marker. Serves as an internal control for normalization in dual-luciferase assay format, providing more accurate interpretation of results
TK-Renilla Control (luc) Thymidine kinase promoter driven Renilla luciferase gene with puromycin selection marker. Serves as an internal control for normalization in dual-luciferase assay format, providing more accurate interpretation of results

Procedure

Cignal Lenti Reporter Control Assays are pretitered, transduction-ready lentivirus particles used for optimizing transduction conditions. Use standard procedures to transduce Cignal Lenti Reporter Control Assays into selected cells, and treat control-transduced cells in the same way as reporter-transduced cells.
