Sensitivity of pigtype Yersinia Ab.
Serum samples from a temporal study of 9 pigs experimentally infected with Yersinia enterocolitica were analyzed with pigtype Yersinia Ab (short protocol). Samples were taken within a 5 week period. From 17 days-post-infection (d.p.i.) to the end of the temporal study (33 d.p.i.) all infected animals tested positive, providing a diagnostic sensitivity for this sample set of > 99%.

Specificity of pigtype Yersinia Ab.
A Yersinia-negative population of 94 serum samples was tested using pigtype Yersinia Ab (short protocol). An S/P ratio (ratio sample/positive control) below 0.3 indicates the sample is negative for antibodies to Yersinia. All samples tested negative, providing a diagnostic specificity for this sample set of 100%.
Performance
pigtype Yersinia Ab is specific for the detection of antibodies to Yersinia spp. A negative population of 94 serum samples was tested. All of the samples tested negative for the Yersinia-specific antibodies, providing a diagnostic specificity for this sample set of 100% (see the figure "Specificity of pigtype Yersinia Ab"). Serum samples of 9 experimentally infected pigs were tested using both the short and the overnight sample incubation protocol. All of the samples tested positive 17 days-post-infection, providing a high diagnostic sensitivity (see the figure Sensitivity of pigtype Yersinia Ab).

Principle

pigtype Yersinia Ab is an ELISA (enzyme-linked immunosorbent assay) with ready-to-use colored reagents. The test plate is coated with inactivated antigen. During the sample incubation, Yersinia-specific antibodies bind to the immobilized antigen. The anti-IgG-HRP conjugate detects antibodies bound to the antigen. The color reaction is started by adding the substrate solution and stopped after 10 minutes. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of anti-Yersinia antibodies in the sample.

Procedure

 

Serum, plasma, and meat juice samples can be tested after dilution.

 

Samples are incubated for 1 hour with the test plate and if antibodies to Y. enterocolitica are present, they will bind to the antigen on the plates. Unbound material is removed by rinsing.

The monoclonal anti-IgG-HRP conjugate is added. Unbound conjugate is rinsed out.

The color reaction is started by adding the TMB Substrate Solution and stopped after 10 minutes. The optical density (OD) is measured using a spectrophotometer. The ratio sample OD to Positive Control OD is calculated from the OD values obtained with the test sample and the Negative Control, which contains no Yersinia-specific antibodies. The values are easy to interpret.
