Performance
flocktype Salmonella Ab is specific for the detection of antibodies to Salmonella enteritidis and Salmonella typhimurium. Tests were run using the flocktype Salmonella Ab Kit in the Deventer International Proficiency Testing Scheme (PTS) ring trial. Sample sets were from either vaccinated chickens (samples 3, 4, and 5) or from infected chickens (samples 2, 6, 7, and 8). Five positive serum samples were correctly scored as being positive for Salmonella antibodies (samples 3, 4, 5, 6, and 8). Due to the early stage of infection, no specific antibodies could be detected in samples 2 and 7. Sample 1 was correctly scored as negative (see the table Deventer ring trial Salmonella 2011 results).

Deventer ring trial Salmonella 2011 results
Sample  Titre  Expected result  flocktype Salmonella Ab Kit result   Origin
1 469 Negative Negative   SPF layers 52 weeks old
 2 401  Positive  Negative   SE field strain 21 d.p.i.
 3 16549  Positive  Positive  TAD ST 28 d.p.v.
 4 15925 Positive Positive  TAD SE 28 d.p.v.
 5 1710 Positive  Positive  Nobilis SG 9R 28 d.p.v.
 6 23618 Positive  Positive  SG field strain 20 d.p.i.
 7 20 Positive Negative   S. Infantis 14 d.p.i.
 8 20858 Positive  Positive  SE field strain 27 d.p.i.

The kit correctly scored 6 of the 8 samples. Due to the early stage of infection, no specific antibodies could be detected in samples 2 and 7.
Principle

flocktype Salmonella Ab is an ELISA (enzyme-linked immunosorbent assay) with ready-to-use colored reagents. The test plate is coated with a Salmonella-LPS antigen mix. During the sample incubation, Salmonella-specific antibodies bind to the immobilized antigen. The anti-IgY-HRP conjugate detects antibodies bound to the antigen. The color reaction is started by adding the substrate solution and stopped after 10 minutes. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of anti-Salmonella antibodies in the sample.

Procedure

Serum and plasma samples can be tested after dilution.

Samples are incubated for 30 minutes with the test plate and if antibodies to Salmonella are present, they will bind to the antigen on the plates. Unbound material is removed by rinsing.

The polyclonal anti-IgY-HRP conjugate is added. Unbound conjugate is rinsed out.

The colour reaction is started by adding the TMB Substrate Solution and stopped after 10 minutes. The optical density (OD) is measured using a spectrophotometer. The results are easy to interpret.

Applications
