Performance
flocktype NDV Ab is specific for the detection of antibodies to Newcastle disease virus in chicken serum samples. Tests were run using the flocktype NDV Ab Kit in the Deventer International Proficiency Testing Scheme (PTS) ring trial. Sample sets were from either vaccinated chickens (samples 1, 2, 3, 4, 5, and 6) or from an infected chicken (sample 8). All positive serum samples were correctly scored as being positive for NDV antibodies (samples 1, 2, 3, 4, 5, 6, and 8). Serum sample 3 was taken from commercial layers that had been vaccinated 3 times with a live NDV vaccine and once with an inactivated NDV vaccine and therefore correctly tested positive. The SPF serum sample 7 correctly tested negative (see the table Deventer ring trial NDV 2007 results).

Principle

Newcastle disease virus (NDV) gives rise to respiratory, circulatory, gastrointestinal, and nervous symptoms in chickens, depending on the age and immune status of the infected chicken and the virulence and tropism of the infecting strain.

flocktype NDV Ab is an ELISA (enzyme-linked immunosorbent assay) with ready-to-use colored reagents. The test plate is coated with a recombinant nucleocapsid protein from the virus. This protein is the major structural protein of NDV.

During the sample incubation, NDV-specific antibodies bind to the immobilized antigen. The anti-IgY-HRP conjugate detects antibodies bound to the antigen. The color reaction is started by adding the substrate solution and stopped after 10 minutes. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of Newcastle disease virus antibodies in the sample.

Procedure

Serum and plasma samples can be tested after dilution.

Samples are incubated for 30 min with the test plate. If antibodies to infectious bursal disease virus are present, they will bind to the antigen on the plates. Unbound material is removed by rinsing.

The anti-IgY-HRP conjugate is added. Unbound conjugate is rinsed out.

The colour reaction is started by adding the TMB Substrate Solution and stopped after 10 minutes. The optical density (OD) is measured using a spectrophotometer. The results are easy to interpret.
