Tests were run using the flocktype Mycoplasma Mg/Ms Ab in the Deventer International Proficiency Testing Scheme (PTS) ring trial. All 6 positive sera were correctly scored as being positive for Mg or Ms antibodies, as a result of infection (samples 1, 4, 5, 7) or vaccination (samples 2 and 3). The Mycoplasma meleagridis positive serum, as well as the Nobilis vaccinated SPF layer, were correctly tested negative. This vaccine does not induce a humoral immunity, and therefore gives negative results in all commercially available ELISAs. flocktype Mycoplasma Mg/Ms Ab is a reliable tool for detecting antibodies to Mg and Ms in poultry populations.

Mycoplasma infections are spread worldwide and cause severe economic losses in poultry farms due to chronic respiratory diseases, decreased growth, and reduced egg production. Morbidity and mortality can vary widely and depend on environmental conditions (e.g., stress, coldness) and secondary infections (other mycoplasma species, bacteria, or viruses).
M. gallisepticum infections can cause chronic respiratory diseases in chicken and sinusitis in turkeys. Infection with M. synoviae leads to subclinical disease of the upper respiratory tract and even to synovitis, tendovaginitis, and bursitis. flocktype Mycoplasma Mg/Ms Ab permits quick and reliable surveillance of poultry flocks for antibodies to Mg and Ms induced by infection or vaccination.
flocktype Mycoplasma Mg/Ms Ab is an indirect ELISA. The microtiter plate is coated with inactivated mycoplasma antigen. During the sample incubation, antibodies specific to Mg or Ms bind to the immobilized antigen. Unbound material is removed by rinsing. Serum antibodies bound to the antigen are detected by the horseradish peroxidase (HRP)-conjugated anti-IgY antibody. Unbound conjugate is removed by rinsing.
A colorimetric reaction is initiated by adding Substrate Solution and stopped after 10 minutes. In the presence of Mg- and Ms-specific antibodies, within the sample, HRP catalyzes a blue color development, which turns yellow after adding the Stop Solution. The optical density (OD) is measured in a spectrophotometer. The OD values correlate with the concentration of Mg- and Ms-specific antibodies in the sample.
