Product Details

The QuickLyse Miniprep Kit provides a rapid spin-column-based method for isolating sequencing-grade plasmid DNA. Bacterial cell resuspension, lysis, and DNA binding are achieved in a single buffer, enabling fast DNA purification with minimal hands-on time.

Performance

The QIAGEN QuickLyse Miniprep Kit uses one-step lysis technology for ultrafast preparation of up to 15 µg plasmid DNA. The quality of the DNA ensures robust performance in applications, such as sequencing, restriction analyses, and cloning (see figure " Reproducible yields").

The QuickLyse Miniprep Kit yields sufficient plasmid DNA for any downstream application, including sequencing (see figure " Long read-lengths in automated sequencing"). Up to 15 µg of high-copy plasmid DNA can be obtained from 1.5 ml of bacterial culture (see figure " High yields of high-quality plasmid DNA"). Isolated plasmid DNA has OD260/280 ratios of 1.7–1.9, indicating that the quality and purity is suitable for all sensitive applications. Sequencing read lengths of over 700 bases are easily achieved.

Principle

Unlike most protocols that use a 3-step lysis procedure, the QuickLyse Miniprep Kit combines enzymatic and osmotic processes to lyse bacterial cells in a single, 3-minute step. In addition, QuickLyse technology uses fewer buffers, simplifying handling and saving time: 24 plasmid DNA minipreps can be prepared in less than 22 minutes.

Procedure

Bacterial cells are resuspended and lysed and DNA is bound in a single buffer, enabling fast DNA purification with minimal hands-on time. After a 3-minute lysis step, the clear lysate is applied directly to a QuickLyse Spin Column. Plasmid DNA binds to the column membrane and is eluted after a single wash step (see flowchart " QuickLyse procedure"). The isolated DNA can be used directly in standard applications, including automated sequencing, PCR, restriction analysis, and cloning.

What is the largest plasmid size that can be purified using the QuickLyse Miniprep Kit?

The QuickLyse Miniprep Kit can be used for plasmids up to 9 kb in size. It is not recommended for Large-Construct purifications.

No, this is not possible since the columns do not fit properly on the QIAVac 24 Plus manifold and the vacuum is not strong enough.
What volume of bacterial culture can be processed using the QuickLyse Miniprep Kit?

Normally, 1.5 ml bacterial culture is used with the QuickLyse Miniprep Kit. Only if the OD600 is < 2.0, up to 3 ml culture may be used. Growth in LB medium is strongly recommended. Growth in nutrient-rich media, such as 2xYT, is not recommended.

How can I increase the DNA yield from low-copy plasmids using the QuickLyse Miniprep Kit?

When using the QuickLyse Miniprep Kit, the addition of 125 µl isopropanol to the lysate, prior to transferring it to the QuickLyse Spin Column, may increase plasmid DNA yield in some cases.

Can the bacterial pellet in the QuickLyse Miniprep procedure be resuspended by pipetting up and down or shortening the vortexing time?

No. Vortexing for 30 seconds as described in the QuickLyse Miniprep Handbook is absolutely critical to resuspend and lyse the bacterial cells. Failure to follow this step will result in greatly reduced DNA yields.

What is the composition of elution buffer QLE in the QuickLyse Miniprep Kit?

Elution Buffer QLE of the QuickLyse Miniprep Kit contains 10 mM Tris-Cl and 0.1 mM EDTA (pH 8.5). Due to the very low concentration of EDTA, enzymatic downstream reactions such as PCR and cycle sequencing are not inhibited.

Yes, nuclease-free water may be used for the elution of plasmid DNA purified with the QuickLyse Miniprep Kit. However, for higher DNA stability and optimal yields, it is recommend to elute using the provided elution buffer (Buffer QLE).
