Product Details

exoRNeasy Serum/Plasma Maxi Kit采用亲和性膜离心柱,可从exosomes和来自血清或血浆中的其他细胞外微泡中高效分离RNA。Maxi规格的分离柱可处理多达4 ml的大体积样本,因此能够可靠检测低丰度RNA。该试剂盒能够快速、便利的获得结果,十分适用于敏感的下游应用。

Performance

exoRNeasy纯化exosomes和其他细胞外微泡与超速离心法相比,不仅速度更快,并且产物纯度更高。扫描电镜显示,虽然这两种纯化技术都能够纯化预期大小的完整的囊泡,但超速离心法的产物还含有很多不符合预期的更小、或其他形状的细胞结构(参见Intact vesicles are eluted from the exoEasy membrane with higher purity compared with ultracentrifugation)。
 
此外,Bioanalyzer分析显示,exoRNeasy和传统超速离心方法分离得到的大RNA和小RNA长度和产量相似(参见exoRNeasy isolates small and large RNAs from extracellular vesicles in plasma)。

通过RT-qPCR实验对exoRNeasy纯化的RNA产物和exoEasy分离柱流出液进行分析,结果显示,exoRNeasy的产物包括血浆中所有mRNA,包括细胞外囊泡的miRNA,且不含细胞(参见exoRNeasy captures all mRNA and vesicle-specific miRNAs in plasma)。

Principle

exoRNeasy Serum/Plasma Maxi Kit改进了传统的超速离心微泡分离方法,能够在20分钟内快速纯化得到细胞外囊泡。该试剂盒采用Exosome Diagnostics, Inc.公司的技术,通过离心柱和特制缓冲液,从多达4 ml的预过滤血浆中分离exosomes。之后使用QIAGEN miRNeasy技术分离总RNA。整个实验流程,从血清/血浆样本到RNA产物,只需1小时。

Procedure

从细胞外微泡分离RNA的完整实验包括两步:纯化exosomes和分离RNA(参见The exoRNeasy Serum/Plasma Maxi Kit workflow — sample to microvesicles to total RNA in just 1 hour)。

在exosomes纯化步骤,预过滤的血浆(不包含大于0.8 μM的微粒)与Buffer XBP混合,加至exoEasay亲和性膜离心柱上,结合到柱上。使用Buffer XWP洗涤结合在膜上的囊泡,之后使用QIAzol裂解。

在RNA分离步骤,在QIAzol洗脱液中加入氯仿,回收水相,将其与乙醇混合。包括miRNA在内的总RNA结合到离心柱上,洗涤三次后洗脱。
