Product Details

exoRNeasy Serum/Plasma Maxi Kit采用亲和性膜离心柱，可从exosomes和来自血清或血浆中的其他细胞外微泡中高效分离RNA。Maxi规格的分离柱可处理多达4 ml的大体积样本，因此能够可靠检测低丰度RNA。该试剂盒能够快速、便利的获得结果，十分适用于敏感的下游应用。

Performance

exoRNeasy纯化exosomes和其他细胞外微泡与超速离心法相比，不仅速度更快，并且产物纯度更高。扫描电镜显示，虽然这两种纯化技术都能够纯化预期大小的完整的囊泡，但超速离心法的产物还含有很多不符合预期的更小、或其他形状的细胞结构（参见Intact vesicles are eluted from the exoEasy membrane with higher purity compared with ultracentrifugation）。

Principle

exoRNeasy Serum/Plasma Maxi Kit改进了传统的超速离心微泡分离方法，能够在20分钟内快速纯化得到细胞外囊泡。该试剂盒采用Exosome Diagnostics, Inc.公司的技术，通过离心柱和特制缓冲液，从多达4 ml的预过滤血浆中分离exosomes。之后使用QIAGEN miRNeasy技术分离总RNA。整个实验流程，从血清/血浆样本到RNA产物，只需1小时。

Procedure

从细胞外微泡分离RNA的完整实验包括两步：纯化exosomes和分离RNA（参见The exoRNeasy Serum/Plasma Maxi Kit workflow — sample to microvesicles to total RNA in just 1 hour）。

在exosomes纯化步骤，预过滤的血浆（不包含大于0.8 μM的微粒）与Buffer XBP混合，加至exoEasay亲和性膜离心柱上，结合到柱上。使用Buffer XWP洗涤结合在膜上的囊泡，之后使用QIAzol裂解。

在RNA分离步骤，在QIAzol洗脱液中加入氯仿，回收水相，将其与乙醇混合。包括miRNA在内的总RNA结合到离心柱上，洗涤三次后洗脱。

Applications

exoRNeasy isolates small and large RNAs from extracellular vesicles present in plasma.

Bioanalyzer sizing was performed with vesicle-derived RNA purified by two methods. The plasma was pre-filtered (0.8 µm) to exclude larger particles and subjected to either ultracentrifugation, the current gold standard of vesicle isolation, or the exoRNeasy procedure. Both methods purify RNA of similar size and yield.

exoRNeasy captures all mRNA and vesicle-specific miRNAs in plasma.

Intact vesicles are eluted from the exoEasy membrane with higher purity compared with ultracentrifugation.
