Genomic DNA was purified from 48 x 350 µl samples (white-cell count 4.9 x106 cells/ml) of human whole blood using the EZ1 DNA Blood 350 µl Kit. Blood samples were taken from 1 individual. Average yields from each run of 6 samples are shown. DNA yield was quantified by absorbance measurements (A260) with background correction. Purified DNA was eluted in 200 µl RNase-free water. Average yield was 8.2 µg (S.D. = 0.23). Average DNA purity (A260/A280) was 1.85 (S.D. = 0.01).|DNA was purified from 200 µl blood samples using the EZ1 DNA Blood 200 µl Kit (upper lanes) and from 350 µl blood samples using the EZ1 DNA Blood 350 µl Kit (lower lanes). Lanes 1–6 and 7–12 are from the first and last of 8 processing runs on the workstation, respectively. M: 1000 bp DNA ladder (100 ng); +: positive control; : negative control. 2 µl (1%) of each eluate was visualized on the agarose gel.|

Principle

使用EZ1 DNA Blood 350 µl Kit进行自动纯化需使用EZ1 Advanced XL和EZ1 Advanced XL DNA Blood Card或EZ1 Advanced XL DNA Buffy Coat Card、EZ1 Advanced和EZ1 Advanced DNA Blood Card或EZ1 Advanced DNA Buffy Coat Card、BioRobot EZ1和EZ1 DNA Blood Card或EZ1 DNA Buffy Coat Card。EZ1卡预装有纯化程序,在控制面板提供操作向导,并提供工作站的运行指令。使用硅胶膜磁珠技术,每批可纯化1–14个样本(EZ1 Advanced XL)或1–6个样本(EZ1 Advanced和BioRobot EZ1)。预封装的试剂条和仪器的自动感控锁可降低污染的风险。

Procedure

将全血样本(350 µl)加载到工作站中,在全自动EZ1 DNA纯化操作后,DNA被一步纯化。基因组DNA洗脱体积为200 µl、100 µl或50 µl,可直接用于各种下游应用。
